The tank flush connection is normally for flushing the black waste tank. Look for it in your manual under the waste tank section. It may be listed as an option as not all RVs have them. You would only use that when you had your sewer hose connection in place and the black waste valve open.
You normally flush out your fresh tank by using the tank fill or fresh water connection, depending upon how your RV is designed. My unit has a diverter valve that directs fresh water connections either to the tank, for filling/flushing, or directly to the plumbing. Consult your owners manual under the winterizing/dewinterizing section for proper procedures.
