My 2000 Winnebago 37G V10 has a Banks 49140 and a transcommand 62570 installed in 2000. Is there any updates that should be done to tweak the system.

These types of systems are not typically update-able. Change your air-filter and fuel filter and perhaps pull the plugs and check those or replace them.

Consider a solution from 5 Star Tuning if you want to pursue a "tweak!"

Brazels RV Performance will also have some tweaks up their sleeve too.
