Whats the best way into the park with a 40' motorhome and toad?

It would depend upon what direction you're coming from and where your reserved RV site is. There's some good info on this website that may answer your question.
RV Camping - Yosemite National Park (U.S. National Park Service)
I personally have never towed anything that big into the park and always try to stay outside in private CGs and take a small vehicle into the park proper.

That's where I got my info. There are 6 campsites each in upper and lower pines campground capable of taking a 40' RV.
