Google maps is your friend...

US 29 to Winchester. Gets complicated beyond there.

Use I-81 and I-87 until you get past DC, NYC and Boston

US 81 to US 90 (toll not too expensive) to I87

I used to live in Essex Junction - traveled that route dozens of times. My preference: 81 north to Binghamton, I-88 to Albany, I-87 north to 149 (exit 20, if memory serves), rt-22, rt-4 into VT and rt-22a up to Vergennes where it connects to rt 7. Fastest way, less traffic after you get out of PA, not bad hills on I-88 and plenty of room for RV's (even on 149). Have done 81 over to 90 but long hills getting to 90 and no faster + tolls. Enjoy Vermont - it's beautiful this time of year!

Doing Essex(~560 miles) in one day? Assuming you are driving a RV, that is pretty ambitious.
