I-75 would be the most direct route, many other roads could be chosen for variety and to avoid traffic at Atlanta, Cincinnati, etc. I hesitate to suggest stops, because I have no idea as to your interests or passengers. Will you have children, young adults along? Pets? Are you interested in history, crafts, shopping, nature, hiking, music, biking? If you use Google to search for things that interest you at any city or region along the way, you'll find a wealth of information and things to visit.
Traveling across the border to Canada doesn't require a passport, but you will need proof of citizenship, papers for your pets, and if you have a conceal/carry permit for a gun, prepare to have your RV searched even if you're smart enough not to try to take a gun into Canada. Read up on border crossing requirements to save yourself time and embarrassment.
