I worked near YNP every summer for 20 years and traveled from Ohio by about every route available. If you come out of YNP through the Tetons (a place you shouldn't miss) US 26 will take you SE across Wyoming. At Casper, you'll get on I-25, but if you follow US 26 when it branches off, you can visit Fort Laramie and Scottsbluff, NE. US 26 joins I-80 near Ogallala, NE.
The other route would be to turn onto US 287 13 miles S of Crowheart, WY. US 287 is good 2 lane road all the way to Rawlins at I-80. On US 287 you will go through Ft. Washakie (Sacagawea grave) and about 12.5 miles S of Jeffrey City you'll see Split Rock, a famous landmark of the Oregon Trail and a Pony Express Station.
