Depends on what is being pulled behind it, your driving speed and style, etc. Nearly impossible to answer.

Both pulling the same load in the same conditions. Which is better

This might be as close as you can get to the answer you look for without more stringent testing standards.

2015 Ram 3500 vs. Ford F-350: Highway MPG Comparison [Video] - The Fast Lane Truck

If the only thing you will look at is the fuel mileage when buying a truck, you may end up disappointed with other things. What other criteria matter to you that maybe we can help you with?

Super broad question...... you haven't even specified gas or diesel. In short, there is no huge difference between brands.

Drive both and see which one you feel more comfortable in. You might not notice a couple mpg difference but you will notice the creature comforts you wanted in the truck you didn't get. I have an f350, but only because to me it was more comfortable than the dodge or Chevy models that were similar. The other two were fine trucks as well just not as comfortable to me. You might find that one of the others is more comfortable to you.

Towing, my guess is there's no more than 1-1.5 mpg difference. Which one is better is anybody's guess. JMO but I'd get the one that doesn't make my butt sore after 8 hrs on the road.

If you are looking at a diesel truck and you are worried about cost of operation on a new truck. Then you need to factor in how much DEF the truck uses between fill ups of DEF, filter cost such as fuel, oil and air. Or just buy the truck that fit's your needs and enjoy.

Real work requires fuel & air mixed in the correct ratio to build horsepower and torque, there is no perpetual motion, yet. W=BTUs
So the engine with the highest HP/LBS-Ft. ratings has the "potential" to use more fuel. Whether it does or not is somewhat determined by the operation of the equipment. Think of them as 400+hp fuel pumps.
In order to produce 500 HP a pump will require a larger volume of fuel+air. it's not magic.

Pick the one that makes you feel better about spending so much.
"So, What's in your wallet?"

Be well.
