A lot depends on two things.. One is the length of hose on the outlet of your regulator, and the position of the regulator and the other is how much trouble do you want to go through.
The regulator connects to the tank valve with a standard POL fitting, the new ACME fittings on the modern tanks are (At least every one I've seen) reverse compatable with the older POL hoses and connectors so you could just unhook from your tank and hook to the portable. however this is not all that easy.
If you install the Extend-a-stay (And please use a quality product, not a el-cheapo home made knockoff) your hook up is way way easier,, Just screw the hose into the inlet, and screw it into or onto the portable tank (Depending on which tank end POL or ACME it has) and open the tank valve.
The Extend-a-stay has all manner of safety check valves and such inside it so there is no danger of back filling the main tank, there is no propane loss via the inlet when you disconnect the portable tank (Just a bit from the hose) and such.
PLUS, you also get the advantages of the Extend-a-flow. This feature is built into the Extend-a-stay as well, This allows you to run a hose to your portable grill or other propane device and run it off the main tank
I have seen some cheap immitations where you had to remember to turn this or that valve and trust me.. Sooner or later.. Either you forget or that valve gets turned by means not fully understood and ... that's dangerous. The check valves in the extend-a-stay make sure this does not happen.
