What Gear

I'm brand new with flat towing. I've got all the lights and brake buddy working and double checked. I've manually disconnected the drive shaft but it just dawned on me that I don't know if it matters what gear I leave the transmission in. I assume neutral but can't find that info anywhere on line. Any help would be appreciated. I'm towing a 1992 Chevy S10 Pickup with a 5 speed manual transmission. Thanks.

If you have driveshaft disconnected, it doesn't matter what gear your in. Personally if auto, I would put in park so you can pull key, unless you have steering lock, in which case you'll have to leave key in. Manual transmission , probably first gear, same on key lock. I'm assuming your towed is two wheel drive. Sorry I see it is a manual trans, biggest issue wheel lock

I tow mine in park. I would tow a manual in 1st gear if for no other reason, you will know right away if the driveshaft disconnect was not fully disengaged.

Do you need to disconnect the driveshaft with a manual trans?

Usually not, but there may be some. Manual trans get lubed by splash from the gears and oil slingers. If the lube is circulated by the engine input then no, they can't be recreationally towed unless the engine is running. But usually the lube is distributed by the output shaft which turns when the driveshaft is connected and the vehicle is towed four down.
