Air bubbles can be tricky. I learned that from bleeding brakes on my airplane. I could see the air bubble and pump plenty of fluid through but the darn bubble persisted. Putting a vacuum on it did the trick but if I could have moved the line to a level place or perhaps lower the bubble would have come out..
The fresh water system drain as shown in the diagram lets you drain the tank. Pushing water into that pipe may push any trapped air out of the pump system. Worth a try at least. You will need to turn the pump on of course.
But OP changed position of the FILL valve and now has a working System....Unless I misinterpreted OPs post
