Most often in a diesel pusher the fill valves will be in front, near the expansion valve. There's also usually a set of schrader valves on the A/C compressor on the engine. I caution doing it yourself, unless you have a full manifold set of gauges. R-134 refrigerant works best when the high and low pressures are within ranges at the ambient temperature. The cans of refrigerant available at Walmart and auto parts stores only have a low pressure gauge. Putting too much in can cause poor cooling, much as too little refrigerant does.
In addition, you wouldn't need to add refrigerant unless you had a leak. Adding dye can help find a leak, but in a DP you've got 70' or more of hoses to look for the red or purple dye. In my opinion, it's worth taking to an auto A/C shop where they can check with a 'sniffer' to find any leaks, fill with the proper amount of refrigerant and lube oil.
