My RV has two 6 volt batteries why can’t I use 1 12 volt battery

I guess that you are talking about coach batteries "house"...
2 X 6 VOLTS batteries will give you more AMPS or a longer time for usage then one single or even 2 X 12 VOLTS batteries !

You can !
That's all you need if your going from campground to campground. If dry camping things change.

You can and many do. A pair of 6v golf car batteries are a very cost-effective wy to get 210-225 amp-hours of battery, but a pair of Group 27 12v's would yield the same.
