Best route for what? You can run almost all the way thru Texarkana, Little Rock, Nashville, Indianapolis, Almost all the way on interstates. If that is your style, go for it. Or you can take almost all back roads and see the USA. All depends on what you want to accomplish. That is 1200 miles or so and you can make it in a few days or a month. Maybe you could be a little more specific about what your wants and needs might be.

Everyone will have opinions but since I've run something close to what you are asking about here is my suggestion.
GOOGLE MAP
As Ron said, it is hard to go much further without an idea of what you are driving, what kind of pace you want to keep and other similar factors that will affect our advice.

Sorry, I should have given more information. We will be driving a 5th wheel and want to drive about 300 to 350 miles a day. We are specifically going to look at new furniture for our trailer as ours is so uncomfortable. We will do sightseeing once we get there and possibly on the way back. We were thinking about all interstates as you said, but wondering how congested Indianapolis is and the best way to tackle going through it.

Thanks for the google map, hadn't thought about that route. We are planning on 300 to 350 miles a day driving a 5th wheel. Any suggestions on campgrounds with your route would be appreciated.

On the go, we have driven some of the route in IL and In. Rt 24 from I-57 east. It's not a bad road at all. We stayed at Cabooselake Campground in Remington. It was fine for an over night.

South to North places we have actually been in.
Hitchin' Post RV Park, Hallsville, Tx
Fairly nice for an over night but we didn't look to see what was around it for a longer stay. Nice laundary facilities and friendly folks.
Catherine's Landing in Arkansas
A very nice resort and great point to visit the Hot Springs area. This is worth a few days to visit the parks and Hot Springs down town.
Downtown Riverside NLR
One of our favorite stops as we transition to/from IL to McAllen TX. Easy access to downtown LR and a walk bridge from it to the Clinton Library. We haven't seen enough of LR yet to get tired of visiting this park.
Hinton RV Park, Sikeston, MO
This is our "MUST" overnight stop. Easy access, nice facilities and home of Lambert's Cafe, home of the Thrown Rolls. Yep...crazy busy restaurant but fun! Lambert's is about 5 minutes away and has a shuttle that will pick you up and the driver will get you straight into the restaurant right past the 1 hour waiting line! Tip appropriately! No need to unhook if you are just overnighting.
Gun Creek on Rend Lake near Benton, IL
We actually stayed at Gun Creek but this is a very nice state/COE CG area as a whole and all have some great spots on or near the lake. Consider South Marcum or Wayne Fitzgerrell SP. These are all places to sit down for a few days and unwind. Fishing is clearly something that can be done here if that is your thing.
Our current home base is D & W LAKE, Champaign, IL.
This is "home" for us and we love it here. Nothing fancy but the owners are fantastic and sites are easy access. If nothing else another nice overnight place with quick access to shopping and restaurants.
Indiana Beach, Monticello, IN
Been there done that...so-so. If you like amusement parks than this might be a nice place but we haven't use those facilities. We have family in that area so we use either of the 2 CGs. Holiday weekends are a ZOO! Not a bad place in the off season but too busy for us during peak.
Shipshewana South CG
We spent a couple nights there having an Amish Cooling unit installed at the factory near by. It's Amish, its got a huge flea market and can be fun. Traffic north of the CG can be CRAZY when the flea market is in session.
