Aside from traffic, you shouldn't have any trouble.

As usual, there are TWO issues here-- OK for the coach and OK for the DRIVER.
Said another way, we can not address whether you are skilled and comfortable with mountain driving.
If your experience is only flat land driving, probably not. Done mountain driving and are familiar with using gear (up and DOWN), go for it.

You are good to go!
I happen to like "technical driving". Others, not so much!

Are you talking about taking Hwy 441 from Knoxville to Sevierville and then TN 66 down to Pigeon Forge, or are you picking up Hwy 321 off of I-40 in Eaton then heading East to Pigeon Forge. If the latter, it sounds like you just want to have some fun driving Hwy 321.

Hwy 321 Knoxville to Pigeon Forge
The easiest way from Knoxville to Pigeon Forge is I-40 east to exit 407 (Route 66), then south to Sevierville then continuing on to Pigeon Forge on U.S. 441 (BTW: 321 does not go through Knoxville - - it runs from I-40 to Gatlinburg & Pigeon Forge and on to Townsend, Maryville & Lenoir City)

There is one steep part with a switchback curve on it. Otherwise, it is a 2 lane road with very little shoulder. If you were in Maryville, you might consider it but if in Knoxville I-40 to TN-66/US-441 is the way to go and it's the way I go living just south of Maryville.
