Your planned route is o.k. for your RV although it's not "mostly flat".
On 285 south of Poncha Springs, CO you'll have 5% for 2 miles on a downhill then 4% for a total of 7 miles.
On 24 you'll have a downhill of 5-6% for 5 miles from Tennessee Pass. Close to I-70 on 24 you'll have 6-7% for 4 miles with curves. This will be the section on your whole drive where you'll have to pay attention.
From Leadville, CO you could also take Hwy 91 to I-70, especially if your destination is the Denver area but there are downhill sections of 4, 5, 6% and 3 miles of 7%.
Again, your RV is fully capable of this beautiful drive. It's a popular RVer's route. It's good you asked about the route so there are no surprises but it's not necessary to avoid the route. You won't be the only RVer doing it.
Being concerned about driving secondary roads keep in mind that interstates have their share of grades, also. I-70 around Hwy 24 will have many miles of 5-7% grades. You're in the mountains and you need to get across. These are your options. No one is going fast on these sections so no need to worry about going slow.
Use your braking wisely and get into lower gear before going down; not in the midst of it.
If you drive mountains I'd highly recommend getting the 'Mountain Directory for Truckers and RVers'. It will give you good detail what to expect ahead. It comes in both the Western and Eastern editions.
We've been on those roads often with our 40' motorhome and we love them. Awesome beauty! Plan to stop and spend some time along this route. Lots to see/do.
