Others have already covered most of my suggestions. I would reconsider placing a closet against the foot of the bed. Its amazing how your feet will seek out that closet wall when you are trying to sleep. Also, I am not sure about just flipping the design around. You will be placing a lot of weight directly on the hitch.

Also, I would seek out an old (vintage) airstream to use as your platform. That will probably have better resale potential when you move on to your second and third versions. :-)

Buy a used toy hauler and modify it to fit your needs. Then you can change it back and sell it or have a TH trailer when your house is done.

Small but usable. Being you have a 26' footprint you are limited. I'd want a living area in it. Perhaps a Murphy bed to make more room in the bedroom area. Put a sofa on the wall and a TV attached to the folded up Murphy bed. Swap the bathroom to the other side and your door issue is fixed.

The biggest things I see are no emergency exits which can be done with emergency egress windows. I agree with moving the kitchen sink against the bathroom wall to keep all of the plumbing in one place. Something that drives me crazy is sitting on the toilet with a wall or door directly in my face. You could move the toilet to the end wall facing towards the front or at least put it in the doorway and leave the door open depending on how close you and your mate are. Best of luck on the build.
