Toad weight vs mpg
How a 1,000 pounds less toad weight could make a difference on mpg ?

Very little, if any. I went from a 2005 Honda Odyssey at about 4,500#'s down to a 2014 CR-V at about 3,500 and didn't notice any difference.

Correct, unless you constantly drive in mountainous terrain, the weight difference has virtually no effect on fuel mileage.

I find that my toad impacts my mpg about -.5 mpg vs no toad. I do use tow mode.

A few thoughts:
1000 lbs on a 54,000 lbs GCWR setup (mine) is 1.8%,
My 400ISL will not notice the difference nor will the Allison 3000 change shift points.
1000 lbs on a 16,000 lb GCWR B-Class Sprinter (or equivalent) is 6.3%
A small diesel or gas engine may downshift more, or even run in a lower gear (not OD).
I would think the gas mileage may change in the second scenario.
Just a thought.

I don't think you'll notice a mileage difference, but will notice a difference climbing hills.
